Deafness diverted

Molecular therapy improves hearing and balance in a mouse model of a hereditary disease that causes deafness and balance disorders.
In humans, Usher syndrome is often caused by a mutation in the gene USH1C that results in the production of a truncated form of the protein harmonin — which normally guides development of the inner ear (pictured) — and leads to impaired hearing.
Jennifer Lentz at Louisiana State University in New Orleans, Michelle Hastings at Rosalind Franklin University in North Chicago, Illinois, and their collaborators injected newborn mice with DNA-like molecules that mask the mutation in the cell's protein-expression machinery, causing normal harmonin to be produced. This stopped the loss of inner-ear cells that respond to sound, improved low- and mid-frequency hearing and reduced behaviours associated with balance impairment such as head-tossing and circling. The effects lasted at least six months. The authors suggest that a similar approach may have therapeutic potential in human congenital deafness.

BOX JELLYFISH


Because most jellyfishes are venomous, they are also called sea wasps or marine stingers. Each tentacle contains more than 100,000 cnidocytes with nematocysts, small needles that inject venom into the victim. The most dangerous species are found in the tropical Indo-Pacific but the Caribbean species can also cause serious injuries. This picture was taken in Guadeloupe (French West Indies) by Dominique Marion, a senior scientist from the Institut de Biologie Structurale in Grenoble, France. Tiny fish trapped in a virtual world provide a window into complex brain connections.

A recently hatched zebrafish is swimming upriver for the first time. Its big round eyes, bulging on the front of its eyelash-sized body, scan the surroundings. Suddenly, it sees the scenery flying forwards as a gentle current pushes it backwards. The fish flicks its tail to try to stay in place. Or so it thinks.
In reality, the baby fish is paralysed and suspended in a water-filled Petri dish by glass pipettes. The dish sits on the stage of a US$100,000 microscope in the corner of a darkened, cluttered laboratory. A film, projected from below, has transported the fish to a virtual world in which moving bands of light and dark simulate passing underwater scenery.
Although the fish doesn't move, the motor neurons that control its tail are firing away, just as if it were swimming. And when fed into a computer, those signals can control the video display, giving the fish nearly every sign that it is swimming normally. All the while, Florian Engert's microscope peers deep into the fish's tiny, translucent brain to watch neurons glow green as they fire (see ‘A river of deceit’).

 City-dwelling sparrows and finches incorporate the butts of smoked cigarettes into their nests, seemingly to ward off parasitic mites.

 Isabel López-Rull and her colleagues at the National Autonomous University of Mexico in Mexico City found that nests of house sparrows (Passer domesticus) and house finches (Carpodacus mexicanus; pictured) with higher levels of cellulose acetate, a component of cigarette butts, had fewer mites. The researchers attached fibres from either smoked or unsmoked cigarette filters to parasite-attracting heat traps and placed them in 27 sparrow and 28 finch nests. Traps bearing fibres from smoked filters, which contain more nicotine than those of unsmoked ones, captured fewer mites, suggesting that nicotine — and perhaps other compounds in cigarettes — repel the parasites.
Birds have long been known to line their nests with vegetation that deters parasites, and the authors suggest that the use of cigarette butts is an urbanized form of this earlier adaptation.

Clifford P. Brangwynne and Anthony A. Hyman celebrate the first book to plausibly suggest how life began.

Aleksandr Oparin (seated) posited that life emerged from compounds in the atmosphere of early Earth.
“No religious or philosophical system, no outstanding thinker ever failed to give this question serious consideration.” So wrote Aleksandr Oparin more than 75 years ago, about the quintessential conundrum of how life self-assembled from inanimate components. The Soviet biochemist’s answer is his book The Origin of Life (1936). Roughly based on a pamphlet he published in 1924, this book is an enormous contribution to our understanding of life’s improbable beginnings. In it, Oparin argues that conditions on early Earth nurtured the synthesis of amino acids and their assembly into protocells.

In the current issue of The EMBO Journal, Joly-Amado et al (2012) describe an obese phenotype in a model of AgRP-ablated mice, and link it to a shift in metabolic profile in efferent tissues such as the
liver, muscle and pancreas.

ASTRONOMY: PICKING OUT A PREDATORY PULSAR

Researchers have used raw computing power to hunt down a 'black widow' pulsar that is evaporating its companion star.
Pulsars are stellar remnants that emit lighthouse-like beams of radiation. They often emit gamma rays, but can usually be spotted only if they also emit easier-to-detect radio waves. However, Holger Pletsch of the Max Planck Institute for Gravitational Physics in Hannover, Germany, and his colleagues found the current pulsar (pictured; circled) through a blind search of data from the Fermi Gamma-ray Space Telescope.
Using computers to analyse huge swathes of raw data, the team picked out the pulsar, which takes 93 minutes to orbit its companion star. This orbital period is the shortest of any binary pulsar of this type yet found.

Stress and the city: Urban decay

 Scientists are testing the idea that the stress of modern city life is a breeding ground for psychosis.

In 1965, health authorities in Camberwell, a bustling quarter of London's southward sprawl, began an unusual tally. They started to keep case records for every person in the area who was diagnosed with schizophrenia, depression, bipolar disorder or any other psychiatric condition. Decades later, when psychiatrists looked back across the data, they saw a surprising trend: the incidence of schizophrenia had more or less doubled, from around 11 per 100,000 inhabitants per year in 1965 to 23 per 100,000 in 1997 — a period when there was no such rise in the general population (J. Boydell et al. Br. J. Psychiatry 182, 4549; 2003). The result raised a question in many researchers' minds: could the stress of city life be increasing the risk of schizophrenia and other mental-health disorders?

In a whirl. Pugh's grand prize-winning entry (which also won the "Deep Space" category) shows the Whirlpool galaxy's spiral arms in sharp detail; trails of light show a smaller, neighboring galaxy being slowly torn apart by the Whirlpool's gravitational pull. 

 M51—also known as the Whirlpool galaxy—is a classic spiral galaxy that scientists have studied for centuries. But this mesmerizing new image of the galaxy has nabbed Australian photographer Martin Pugh the top prize in the fourth annual Astronomy Photographer of the Year awards, announced this week. The sharp detail of the spiral’s arms, defined by dark, dusty areas and bright, pink clouds of hydrogen, impressed the judges. “This is arguably one of the finest images of M51 ever taken by an amateur astronomer,” said judge and astronomer Will Gater in a press release. Pugh’s photograph—which was chosen from more than 800 entries from around the world and earned him £1500—as well as those of the winners in categories such as ”Young Astronomy Photographer of the Year,” “Earth and Space,” and “Our Solar System,” are now on display at the Royal Observatory Greenwich in the United Kingdom. Other standout entries include a photograph of the Pleiades cluster, taken by a 15-year-old astronomer, and an image of this year’s transit of Venus across the sun, which was taken on 6 June.

If you're trying to avoid getting pregnant, here's another reason to mistrust the rhythm method of birth control: New research confirms that the fluid in semen, long dismissed as primarily a vehicle for sperm, contains a substance that can trigger ovulation and other pregnancy-supporting hormonal responses in female mammals. The find could lead to new fertility treatments in humans.

Abrupt changes in Greenland ice cycles


An analysis of aerial photographs suggests that ice loss in northwestern Greenland results from discrete events driven by changes in ocean and atmospheric temperature. Kurt Kjær at the University of Copenhagen's Natural History Museum and his team used photographs dating back to 1985 to create a three-dimensional model of ice loss and gain. Rather than exhibiting a uniform melt rate, the model revealed that ice loss peaked in two periods: 1985–93 and 2005–10. The researchers linked this ice loss to warmer oceans and higher summer air temperatures. Ice-sheet models must account for such variability if they are to produce reliable forecasts.

Metabolic support for neurons

The myelin sheath around nerve fibres serves to speed up electrical nerve signals. But it turns out that it also supplies neurons with fuel to support their high metabolic activity


Blood glucose (orange) can be taken up by non-neuronal brain cells known as oligodendrocytes and astrocytes. Inside the cells, glucose is broken down to lactate (blue), which can then be transferred to neurons and used to generate metabolic energy in the form of ATP molecules (yellow). Lee et al.2 show that oligodendrocytes supply nerve fibres (axons) with lactate delivered through a transporter protein (MCT1, not shown) located in the myelin, a membranous sheath around the axons. Lactate can also be produced in astrocytes and then transferred to axons by means of small pores (gap junctions, not shown) through the myelin. In addition, blood lactate may reach neurons through astrocytes and oligodendrocytes.

 Humans evolved to run. This helps to explain our athletic capacity and our susceptibility to modern diseases, argue Timothy Noakes and Michael Spedding.

The forthcoming Olympics in London will celebrate the performance capacity of humans and our remarkable ability to prepare our bodies and minds for specific tasks. But, at the same time as we are pushing our bodies to new limits in athleticism, we are experiencing unprecedented levels of relatively modern diseases such as obesity, diabetes and psychiatric and neurodegenerative disorders.



Although some research has yielded mixed results about the benefits of exercise for the brain5, the literature in general shows that regular exercise can have many cognitive benefits. Some research suggests that exercise has antidepressant effects, at least against mild depression, and may even offset Alzheimer's disease: an exercise programme has been shown to increase the volume of the brain's hippocampus. More data are needed to obtain a clearer picture of the effects of exercise on cognition. Ongoing clinical trials are testing the effects of exercise on Alzheimer's disease and schizophrenia.

MEXICO CITY

Mexican scientists have watched with dismay as their country, Latin America's second-largest economy, has slipped down the research-spending ranks in recent years. Candidates in this week's presidential election have pledged to change that.
Back in 2000, presidential candidate Vicente Fox promised to raise Mexico's gross expenditure on research and development from about 0.4% to 1% of gross domestic product (GERD per GDP, often known as research intensity). Fox won, but despite legislation meant to guarantee the increase, Mexico's GERD per GDP has remained below 0.5%. Science has barely featured in campaigns for subsequent elections, presidential or otherwise.
The attention to research investment comes as Mexico faces competition from other emerging economies. “Brazil has almost caught up with Mexico in GDP per capita, in less than a generation,” says Andrew Selee, director of the Mexico Institute at the Woodrow Wilson Center in Washington DC. “Mexicans are starting to ask why.”
For much of the twentieth century, Mexico was considered to be the leader in Latin American science. Yet although the country still has pockets of research excellence, Mexico's GERD per GDP now ranks among the very lowest in the world's top 40 economies (see 'Paltry pesos').
Brazil, by contrast, has consistently spent more than 1% of its GDP on research since 2006. Its gross expenditure on science, including industrial research, totals almost US$20 billion per year — roughly five times Mexico's tally — despite cuts this year of almost 20% to the US$3.8-billion budget of the Brazilian Ministry of Science, Technology and Innovation (L. Amorim Science and Development Network 6 March 2012).
“Knowledge, research, development and innovation have not been a priority for Mexican decision-makers,” says Juan Pedro Laclette, head of the Scientific and Technological Consultative Forum, a leading think-tank for science based in Mexico City. “If you plant peanuts, you are going to harvest peanuts. What Mexican politicians have planted — have invested — is peanuts.”
The presidential candidate who talks the most about reviving research is Andrés Manuel López Obrador, who leads the left-wing Party of the Democratic Revolution. Polls put him just ahead of Josefina Vázquez Mota of the right-leaning National Action Party, which has held the presidency since 2000, but behind Enrique Peña Nieto of the left-wing Institutional Revolutionary Party, which controlled the Mexican government for more than seven decades until 2000, and became a symbol of corruption and voter fraud.
In the most recent presidential election, in 2006, Obrador came second by such a narrow margin that he held regular protests for five months, during which time he named himself president and even set up an alternative government — machinations that have damaged his credibility this time around. His party, however, has deep roots in academia and enjoys wide support from scientists. René Drucker Colín, a neuropathologist and former coordinator of basic science at the National Autonomous University of Mexico in Mexico City, one of Latin America's biggest universities, is among the researchers who have taken to the campaign trail on Obrador's behalf.
Obrador plans to remedy what many researchers complain is a major handicap for Mexican science: the lack of a clear national research strategy with long-term commitments to nurturing particular fields. He has pledged to boost spending and promote the country's biggest science funder, the National Council on Science and Technology, to a fully fledged ministry — with Drucker Colín at its head — that would control the nation's entire science budget.
Peña Nieto, the current presidential front runner, has said less publicly about science. But in an e-mail to Nature, he wrote: “We need to create a National System for Innovation that promotes it through effective stimuli, intellectual property rights protection, better access to credit and coordination between the public and private sectors.”
Industry contributes less than half of Mexico's GERD per GDP, and the country's scientists, including Pedro Laclette, have long bemoaned the schism between industry and academic research. In the future, wrote Peña Nieto, “private funding will be instrumental to the success of the system”.
Whoever wins the election, researchers are adamant that the time for broken promises is past. “If you take the difference between what they give and what they were supposed to be giving, they owe science a huge amount of money,” says Franco López.

Hydrogen 'Bridge' Connects Two Galaxies

ANCHORAGE, ALASKA—Two large neighbors of our own Milky Way galaxy—Andromeda (upper right) and Triangulum (lower left)—experienced a close encounter in the distant past. When they brushed past each other billions of years ago, cold hydrogen gas was pulled out of the galaxies by their mutual gravity. The faint radio emission of the resulting hydrogen "bridge," the red material stretching from Andromeda in the direction of Triangulum, was first hypothesized based on observations from a Dutch radio telescope in 2004 and has now been mapped in detail by the Green Bank Telescope in West Virginia. The bridge, described here yesterday at the 220th meeting of the American Astronomical Society, extends over hundreds of thousands of light-years, and the hydrogen gas appears to be clumped into clouds a few thousand light-years across. Next time, the two galaxies may not be so lucky. Recent Hubble Space Telescope observations have revealed that Andromeda will collide and merge with our Milky Way in a few billion years. By then, the smaller Triangulum galaxy may have already collided with either of its two larger neighbors. Ultimately, the trio will meld into one giant elliptical galaxy, flinging stars and planets in all possible directions.

Venus crosses the Sun Venus made its transit across the Sun on 5–6 June, watched eagerly by professional and amateur astronomers, as well as by millions of people following it on live webcasts. The rare event occurs twice in close succession roughly every 120 years; the last transit was in 2004 and the next one will not be until 2117. Scientists hoped to get a once-in-a-lifetime snapshot of the climate of the entire planet (rather than the time- and space-limited atmospheric snapshots taken by the Venus Express probe), and to use the transit to check the way we measure exoplanets circling distant stars. Can coral cope with climate change?



Climate change is likely to alter the species composition of coral reefs, rather than wipe out entire reef ecosystems.
Terry Hughes at James Cook University in Townsville, Australia, and his team sampled 132 sites along the full length of the Great Barrier Reef (pictured), spanning 13° of latitude and a range of sea surface temperatures. Of the 12 coral taxa sampled, 11 showed significant differences in abundance across the reef, regardless of how susceptible they were to thermal stress and bleaching. These differences in abundance did not follow changes in latitude or temperature.
This flexibility may enable coral reefs to continue functioning as the environment alters with climate change.
